I've got 2 video cards in the system.

Identical GeForce 8600GT cards.
Each has a DVI port & a d-sub port.

I've got two monitors - one is plugged in HDMI port (it doesn't have a DVI port)...and a HDMI to DVI adapter plugs into the video card.

second monitor - unless I use the d-sub connection, will not turn on... the power button changes to green when I restart the computer, but then says 'no signal'...

I switched the cables around - which ever is plugged into the 2nd video card - won't connect except with the D-sub.

I've updated drivers....the pc sees both video cards... just won't connect on the dvi cable..

Do you have the nVidia control panel installed? You can try the "identify monitors" button to see if that will wake it up. May also be a setting for auto detect that has gone wrong. Is there an onboard video connection on the MB? Is it disabled?

Can't think of anything else off the top of my head... Which OS?

I'll check the onboard video.
