Its a little on the pricey side but it's of the highest quality and I dont mind paying a little more for something thats gonna last.
My partner and I both use a Tumi backpack.
It fits comfortably: Canon 5D and Canon 50D with lenses attached (both wrapped), 2 speedlites, 70-200 lens,
3 pocketwizards, snoot, cables, wire trigger, tripod connector, (16) AA batteries and 2 chargers, (3) canon batteries and charger,
business cards, pens, gels, and my laptop.

It also doesn't scream "I'm a camera bag! Come steal me!"

I have a black one which is cool, but my partner's is BRIGHT yellow which really stands out if someones running away with your bag in the airport, then you can yell "stop the guy with the yellow bag!" or "That's my yellow bag!"

I'm using a Kata DR-467 and love it. Top section will handle a D700 w 24-70. Bottom compartment will hold 3 medium sized lens, or a 7--200 and an SB-600. Plenty of pockets and a laptop pouch. You can put more gear in it than you would want to carry all day. (Ask me how I know...)

Good straps, quality zippers, good material, yellow interior (harder to lose stuff) and a sleeve to fit over the handle of your roller carry-on. Well padded and holds up well to use. And it fits comfortably under the seat in a standard airliner. I'd buy another in a minute.

I got a good deal doing a price match on eBAGS.

The Think Tank stuff is very nice, but pricey (in my opinion). If I were going to use it every day, I'd go Think Tank. I have LowePro stuff I like and use regularly, but have no experience with their backpacks. My other big case is a Pelican 1510, but that's a different class.

check out Crumpler backpacks. Buncha different sizes and styles and they don't look like camera bags. Really nice stuff.
