Like Andy is saying I have no problem playing 5DMKII footage straight from the CF card very smoothly on any of my 3 Mac's I don't own a PC but I heard it's hard to play it. I use Kingston 133X and SanDisk Extreme III CF cards with no problems at all.
I edit in FCP and what I do is change the original 5DII h264/.mov files to apple proress 422 HD codec cause editing the original footage can be a pain on the a$$ even on my 6 month old macpro 8core/16GBram. I edit and render the original timeline to a lossless QT movie then from that original footage using compressor I can compress it to h264 for blu-ray disc or MPEG2 for a regular DVD.
When I am using my macbook 13" I use Premiere Pro CS4 and what I do is very simple, since I know I can't edit HD footage with a dual core machine, what I do is opening Adobe Media Encoder CS4 and compress my original files to a SD H264 codec and then open my footage in premiere pro and presto.
For right now and till the new firmware upgrade for the 5DMKII comes out, when I need to convert my footage from 30fps to a more movie like 24fps I'll do it with cinema tools. It does a great job and it looks and feels very nice.

I use magic video converter, you can specify your video bit rate, size and all that good stuff. I even use this software to convert some of my DVDs, movie clips to use on the iphone, very useful piece of software for cheap.

My windoze quadcore plays anything you throw at it just fine. 7D & 5DII videos are also smooth, time to upgrade for some of you I think
